愈疮散对冰醋酸诱导复发性口腔溃疡大鼠黏膜损伤及组织内炎性因子的影响

摘要:

目的 探讨愈疮散对冰醋酸诱导复发性口腔溃疡(ROU)大鼠黏膜组织损伤的影响及对黏膜组织内肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)浓度的影响。方法 将雌雄各半SPF级健康SD大鼠48只,采用随机数字表法分为空白组8只,造模组40只。造模组采用冰醋酸诱导法制备ROU模型,再随机分为模型组、壳聚糖组、愈疮散低、中、高剂量组(壳聚糖组和愈疮散低、中、高剂量组合称用药组),每组8只。壳聚糖组予口腔溃疡凝胶治疗,愈疮散各剂量组予不同剂量愈疮散治疗。连续用药7 d后,采用HE染色观察各组大鼠溃疡黏膜组织病理改变,采用免疫组化染色观察组织内TNF-α,IL-6,IL-1β的表达,通过酶联免疫吸附法检测组织匀浆内TNF-α、IL-6、IL-1β浓度,采用蛋白质印迹法检测溃疡组织内TNF-α、IL-6、IL-1β的表达。结果 HE染色结果显示,各用药组大鼠溃疡黏膜组织病理损伤均缓解;免疫组化染色结果显示,与模型组比较,各用药组大鼠溃疡组织TNF-α、IL-6、IL-1β表达减少(均P<0.05);酶联免疫吸附法检测结果显示,与模型组比较,各用药组大鼠组织TNF-α、IL-6、IL-1β浓度降低,差异均有统计学意义(均P<0.05);蛋白质印迹法检测结果显示,与模型组比较,各用药组大鼠组织内TNF-α、IL-6、IL-1β表达减少,差异均有统计学意义(均P<0.05);用药组组间比较,差异无统计学意义(均P>0.05)。结论 愈疮散能促进ROU大鼠口腔溃疡黏膜组织的修复,这种作用可能通过降低溃疡黏膜组织内TNF-α,IL-6,IL-1β的浓度、减轻组织内的炎症反应来实现。

Abstract:

Objective To investigate the effect of Yuchuang Powder on mucosal tissue damage and the concentrations of tumor necrosis factor-alpha(TNF-α), interleukin-1β(IL-1β), and interleukin-6(IL-6) in rats with recurrent oral ulcers(ROU) induced by acetic acid. Methods 48 healthy SPF-grade SD rats(equal numbers of males and females) were randomly divided into the blank control group(n=8) and the modeling group(n=40) using a random number table. The modeling group was used to establish the ROU model by acetic acid induction, and then randomly divided into the model group, chitosan group, low-dose Yuchuang Powder group, medium-dose Yuchuang Powder group, and high-dose Yuchuang Powder group, with 8 rats in each group. The chitosan group was treated with oral ulcer gel, while the Yuchuang Powder groups were treated with different doses of Yuchuang Powder. After 7 consecutive days of medication, mucosal tissue pathology was assessed by hematoxylin and eosin(HE) staining, expression levels of TNF-α, IL-6, and IL-1β in tissue were evaluated by immunohistochemistry, concentrations of TNF-α, IL-6, and IL-1β in tissue homogenates were measured by enzyme-linked immunosorbent assay(ELISA), and protein expression levels of TNF-α, IL-6, and IL-1β in ulcer tissue were analyzed using Western blotting. Results HE staining showed that mucosal tissue damage in the medication groups was significantly alleviated compared with the model group Immunohistochemistry indicated reduced expression of TNF-α, IL-6, and IL-1β in the medication groups compared with the model group(all P<0.05). ELISA results showed significantly lower concentrations of TNF-α, IL-6, and IL-1β in the medication groups(all P<0.05). Western blotting confirmed decreased protein expression of TNF-α, IL-6, and IL-1β in the medication groups(all P<0.05). No statistically significant differences were observed among the medication groups(all P>0.05). Conclusion Yuchuang Powder can promote the mucosa lrepair in ROU rats, and this effect may be achieved by reducing the concentrations of TNF-α, IL-6, and IL-1β in ulcer mucosal tissues and alleviating local inflammatory response.
