基于数据挖掘探讨针药联合方案对模拟失重大鼠心功能下降的影响

摘要:

目的 探究针药联合提升心功能的用药选穴规律,验证穴位电刺激联合龙归养心丸对模拟失重大鼠心功能的协同干预作用。方法 文献分析:检索中国知网、维普、万方数据库,筛选2016—2021年所有提升心功能的针灸、中药处方相关文献,通过Excel、Modeler 18.0及Gephi进行频次、配伍关联及复杂网络分析。动物实验:将25只SPF级健康雄性Wistar大鼠分为空白组、模型组、电刺激组、中药组、针药联合组,每组5只,尾悬吊模拟失重28 d进行造模。各组进行相应干预治疗后,采用超声检测心功能指标左心室射血分数、左心室缩短分数。取心肌组织进行HE染色、Hoechst染色、免疫组化检测、蛋白质印迹法及酶联免疫吸附法检测,分析心肌形态、凋亡、胱天蛋白酶3的表达及琥珀酸脱氢酶、乳酸脱氢酶活性。结果 文献分析显示:针药联合以内关穴配伍益气活血中药常见。动物实验显示:针药联合组左心室射血分数和左心室缩短分数升高,显著优于电刺激组和中药组(均P<0.05),且心肌细胞形态损伤明显改善。与模型组相比,电刺激组及针药联合组胱天蛋白酶3表达量降低(均P<0.05)。电刺激组、中药组与针药联合组琥珀酸脱氢酶、乳酸脱氢酶活性较模型组显著升高(均P<0.05)。结论 针药联合可有效提升心功能,龙归养心丸联合穴位电刺激可显著改善失重大鼠的心功能及心血管系统紊乱,穴位电刺激可抑制细胞凋亡,其机制可能与调控胱天蛋白酶3的表达及酶活性有关。

Abstract:

Objective To explore the laws of herb selection and acupoint prescription in acupuncture-medicine combination for improving cardiac function, and to verify the synergistic intervention effect of electroacupuncture stimulation combined with Longgui Yangxin Pill on cardiac function in rats under simulated microgravity condition. Methods Literature on acupuncture and Chinese herbal prescriptions for improving cardiac function published between 2016 and 2021 was retrieved from CNKI, VIP, and Wanfang databases. Frequency, compatibility association, and complex network analyses were performed using Excel, Modeler 18.0, and Gephi. 25 SPF-grade healthy male Wistar rats were divided into the blank control group, model group, electrical stimulation group, medication group, and acupuncture-medication combination group, with five rats in each group. A tail suspension-induced simulated microgravity model was established over 28 days. After intervention, cardiac function indicators including left ventricular ejection fraction(LVEF) and left ventricular fractional shortening(LVFS) were measured using echocardiography. Myocardial tissues were collected for HE staining, Hoechst staining, immunohistochemical detection, Western blotting, and enzyme-linked immunosorbent assay(ELISA) to analyze myocardial morphology, apoptosis, caspase-3 expression, and the activities of succinate dehydrogenase(SDH) and lactate dehydrogenase(LDH). Results Literature analysis showed that the combination of Neiguan(PC6) acupoint with Qi-invigorating and blood-activating Chinese herbs was common in acupuncture-medication combination therapy. Animal experiments revealed that the acupuncture-medication combination group had increased LVEF and LVFS, which were significantly superior to both the electrical stimulation group and the medication group(all P<0.05), with markedly improved myocardial cell morphology. Compared to the model group, caspase-3 expression decreased in both the electrical stimulation group and the acupuncture-medication combination group(all P<0.05). SDH and LDH activities were significantly improved in the electrical stimulation group, medication group, and acupuncture-medication combination group compared to the model group(all P<0.05). Conclusion The combination of acupuncture and medication can effectively enhance cardiac function. Longgui Yangxin Pills combined with acupoint electrical stimulation can significantly improve the cardiac disfunction and cardiovascular disorders in rats under simulated microgravity. Acupoint electrical stimulation can inhibit cell apoptosis. The mechanism may be related to the regulation of Caspase-3 expression and enzyme activities.
